چکیده

In this paper, we report the circular dichroism (CD) spectra of two types of LH2-only mutants of Rhodobacter sphaeroides. In the first, only the wild type LH2 is present, while i the second, the B800 binding site of LH2 has been either destabilized or removed. For the first time, we have identified a band in the CD spectrum of LH2, located at approximately 780 nm, that can be ascribed to the high exciton component of the B850 band. The experimental spectra have been modeled by theoretical calculations. On this basis, the average interaction strength between the monomers in the B850 ring can be estimated to be approximately 300 cm-1. In addition, we suggest that in LH2 of Rb. sphaeroides the angles made by the Qy transitions of the B850 BChls with respect to the plane of the ring are slightly different from those calculated from the crystal structure of the Rhodopseudomonas acidophila LH2 complex.

Exciton (De)Localization in the LH2 Antenna of Rhodobacter sphaeroides As Revealed by Relative Difference Absorption Measurements of the LH2 Antenna and the B820 Subunit

We report the results of relative difference absorption measurements for the LH2 antenna of Rhodobacter sphaeroides and the B820 subunit of Rhodospirillum rubrum at room temperature. It is shown that significant differences between shapes and amplitudes of photoinduced, absorption changes reflect a different degree of exciton delocalization in the intact antenna compared with the dimeric subuni...

Effect of oxygen on levels of mRNA coding for reaction-centre and light-harvesting polypeptides of Rhodobacter sphaeroides.

The relative levels of mRNA for the reaction-centre L and M subunits, B875 (LH1) alpha and beta polypeptides and B800-850 (LH2) alpha and beta polypeptides, have been measured during pigment induction of Rhodobacter sphaeroides. Over the 6 h of the experiment, bacteriochlorophyll levels increased by at least 100-fold. No transcripts for photosynthetic components were detectable at the start of ...

Theoretical prediction of spectral and optical properties of bacteriochlorophylls in thermally disordered LH2 antenna complexes.

A general approach for calculating spectral and optical properties of pigment-protein complexes of known atomic structure is presented. The method, that combines molecular dynamics simulations, quantum chemistry calculations, and statistical mechanical modeling, is demonstrated by calculating the absorption and circular dichroism spectra of the B800-B850 bacteriochlorophylls of the LH2 antenna ...

The mechanism of energy transfer in the antenna of photosynthetic purple bacteria

The mechanism of energy transfer in the antenna system of purple bacteria is investigated by combination of photon echo spectroscopy and disordered exciton theory. In the B800 component of light harvesting complex 2 (LH2), a picture of incoherent hopping between monomers provides an excellent description of the photon echo data recorded as a function of excitation wavelength. In the B850 pigmen...

Uphill energy transfer in LH2-containing purple bacteria at room temperature

Uphill energy transfer in the LH2-containing purple bacteria Rhodopseudomonas acidophila, Rhodopseudomonas palustris, Rhodobacter sphaeroides, Chromatium vinosum and Chromatium purpuratum was studied by stationary fluorescence spectroscopy at room temperature upon selective excitation of the B800 pigments of LH2 and the B880 pigments of LH1 at 803 nm and 900 nm, respectively. The resulting fluo...
